Abstract

A restorative therapy for a disease such as Parkinson's, where substantial cellular degeneration has already occurred at the time of diagnosis, is likely to require a more complex intervention than simply enhancing dopamine output in the brain. Here we assess the value and feasibility of a variety of strategies aimed at either replacing the lost neuronal circuitry or providing one-off delivery of corrective and/or neuroprotective genes.
